According to NFL.com’s Ian Rapoport, the NFL will announce its 2016 regular season schedule on Thursday, which will occur at 8 PM EST:
The @NFL schedule will be announced tomorrow, per an announcement to add to the earlier announcement about a forthcoming announcement

Besides their divisional opponents (the Houston Texans, Jacksonville Jaguars, and Tennessee Titans), the Colts will also play the Kansas City Chiefs, San Diego Chargers, Chicago Bears, Detroit Lions, and Pittsburgh Steelers at home in the friendly confines of Lucas Oil Stadium.
Likewise, on the road and beyond the AFC South, the Colts will travel to face the Denver Broncos, Oakland Raiders, Green Bay Packers, Minnesota Vikings, and New York Jets–with one wildcard:
NFL schedule released at 8 tomorrow night. We already know one game: Colts vs. Jags October 2 in London.

The Colts Week 4 divisional matchup with the Jacksonville Jaguars in London will count as a “road” game, so that has already been determined.
Right now, it’s just a matter of when the rest of the team’s games will exactly occur. It’s sort of like the NFL’s version of Christmas morning.
